Skin cancer is the deadliest type of malignant disease and causes primary mortality worldwide. Dioscin, which exists in medicinal plants, has potent anticancer effects. However, its effects on skin cancer remain unknown. In the present study, the activity and mechanism of dioscin on the human skin cancer A431 cell line were investigated, MTT, colony formation, Transwell, wound-healing, TUNEL, Comet, immunofluorescence and western blot assays were used to assess the effects of dioscin on A431 cells. The results of MTT, colony formation, Transwell and wound-healing assays revealed that dioscin suppressed proliferation, colony formation and invasion of the cancer cells. TUNEL and comet assays demonstrated that dioscin exhibited significant effects on cell apoptosis and DNA damage. Investigations into the mechanism revealed that the expression levels of phosphorylated Ataxia telangiectasia-mutated (ATM) were considerably activated by dioscin, which significantly upregulated the expression levels of p53 to activate mitochondrial apoptosis signaling. Furthermore, the expression levels of BAX, cleaved caspase-3/9 and cleaved poly (ADP-ribose) polymerase were upregulated, and the expression levels of BCL-2 were downregulated by dioscin. Additionally, dioscin markedly downregulated the expression levels of matrix metalloproteinase 2 (MMP2), MMP9, RHO and cdc42, which are all associated with tumor invasion. In addition, p53-small interfering RNA transfection experiments indicated that dioscin exhibited excellent activity against skin cancer in vitro by decreasing p53 expression. Overall, the present results suggested that dioscin inhibited skin cancer cell proliferation via adjusting ATM/p53-mediated cell apoptosis, migration and DNA damage, which should be considered as a potential option for future treatments of skin cancer. 相似文献

Discoid lupus erythematosus (DLE) is the most common skin manifestation of lupus; however, the molecular mechanisms underlying DLE remain unknown. Therefore, we aimed to identify key differentially expressed genes (DEGs) in discoid lupus skin and investigate their potential pathways.To identify candidate genes involved in the occurrence and development of the disease, we downloaded the microarray datasets and GSE52471 from the Gene Expression Database (GEO). DEGs between discoid lupus skin and normal controls were selected using the GEO2R tool and Venn diagram software ( GSE72535http://bioinformatics.psb.ugent.be/webtools/Venn/). The Database for Annotation, Visualization, and Integrated Discovery (DAVID), Enrichr, and Cytoscape ClueGo were used to analyze the Kyoto Encyclopedia of Gene and Genome pathways and gene ontology. Protein-protein interactions (PPIs) of these DEGs were further assessed using the Search Tool for the Retrieval Interacting Genes version 10.0.Seventy three DEGs were co-expressed in both datasets. DEGs were predominantly upregulated in receptor signaling pathways of the immune response. In the PPI network, 69 upregulated genes were selected. Furthermore, 4 genes (CXCL10, ISG15, IFIH1, and IRF7) were found to be significantly upregulated in the RIG-I-like receptor signaling pathway, from analysis of Enrichr and Cytoscape ClueGo.The results of this study may provide new insights into the potential molecular mechanisms of DLE. 目的研究异牡荆素(ISO)对非小细胞性肺癌(NSCLC)细胞的影响和潜在的机制。方法将A549和H1650细胞分别分为空白组、低剂量实验组(4μmol·L-1 ISO)和高剂量实验组(16μmol·L-1 ISO)。用噻唑蓝法检测NSCLC细胞活性,用肿瘤球形成实验检测NSCLC细胞的细胞球形成率,用Western blot法检测NSCLC细胞凋亡、自我更新、无翅型MMTV整合位点家族/β-连环蛋白(Wnt/β-catenin)信号通路相关蛋白的表达水平。结果与空白组比较,低、高剂量实验组中A549和H1650细胞在24,48和72 h的细胞活性均显著降低。低、高剂量实验组和空白组中A549细胞的细胞球形成率分别为(4.18±0.45)%,(2.01±0.67)%和(6.02±0.57)%,切割的半胱氨酸蛋白酶-3相对表达量分别为0.24±0.08,1.25±0.13和0.06±0.07,SRY相关高迁移率族盒蛋白-2相对表达量分别为0.49±0.04,0.25±0.03和1.00±0.09,Kruppel样因子4相对表达量分别为0.68±0.04,0.44±0.03和1.01±0.06,Wnt1相对表达量分别为0.63±0.06,0.28±0.04和1.00±0.06,β-catenin相对表达量分别为0.41±0.05,0.22±0.03和1.01±0.09;与空白组比较,低、高剂量实验组中A549细胞的上述指标的差异均有统计学意义(P<0.05,P<0.01)。上述3组中H1650细胞的上述指标也呈现一致的现象。结论ISO可能通过抑制Wnt/β-catenin信号通路来抑制NSCLC细胞活性和自我更新,并促进其凋亡。 相似文献

目的 研究刺槐素对干眼症(DED)大鼠的保护作用及可能的分子机制。方法 60只SD大鼠随机分为空白组(不做处理);模型组;0.1% SH组;刺槐素0.1%、0.25%、0.5%组。每组10只,除正常组外,每日4次皮下注射12.5 mg/天氢溴酸东莨菪碱(SCOP),共7天。所有的药物均为在眼球表面局部给药,每只眼睛20 μL,每天4次。通过泪液分泌实验检测各组大鼠泪液分泌情况,通过角膜荧光染色检测各组大鼠角膜表面缺损情况,过碘酸雪夫试剂(PAS)染色观察结膜杯状细胞数量,酶联免疫吸附法(ELISA)检测各组大鼠角膜组织中炎症反应和氧化应激相关指标含量。免疫印迹法(Western blot)检测各组大鼠角膜组织中Toll样受体4(TLR4)通路相关蛋白TLR4、髓样分化因子88(MyD88)、核因子κB p65(NF-κB p65)、Pyrin域蛋白3(NLRP3)蛋白表达。结果 与正常组相比,模型组大鼠泪液分泌量、结膜杯状细胞数量明显减少,角膜荧光素染色评分显著升高,但角膜新血管生成评分减少,同时角膜组织中白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)、IL-10、丙二醛(MDA)水平上调,而超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)含量则下调,TLR4、MyD88、NLRP3、p-NF-κB p65蛋白相对表达量均升高(P<0.05);与模型组相比,经刺槐素0.25%、0.5%组治疗后,大鼠泪液分泌量、结膜杯状细胞数量明显增加,角膜荧光素染色评分显著降低,角膜新血管生成评分显著增加,同时角膜组织中IL-1β、TNF-α、IL-10、MDA水平下调,而SOD和GSH-Px含量则上调(P<0.05),尤其是刺槐素0.5%组上述指标变化更明显,而且角膜组织中TLR4、MyD88、NLRP3、p-NF-κB p65蛋白相对表达量较模型组显著降低(P<0.05)。结论 刺槐素对干眼症大鼠具有保护作用,其保护机制与抑制干眼症大鼠的氧化应激反应、炎症反应和TLR4/MyD88/NLRP3通路活性有关。 相似文献

Cutaneous histiocytoses constitute a heterogeneous group of diseases characterised by the cutaneous accumulation of cells with the cytological and phenotypic features of macrophages or dendritic cells. The clinical spectrum ranges from self-resolving, skin-limited conditions to severe, multiorgan disease with a high morbidity rate. Until recently, cutaneous histiocytoses were classified according to the immunophenotype of the pathological cells, with differentiation between Langerhans cell histiocytosis (LCH) [CD1a+, CD207 (langerin)+] and non-Langerhans cell histiocytosis (CD68+, CD163+, CD1a?, CD207?). Over the last 12 years, a number of new pathophysiological findings (in particular, molecular pathology results) regarding histiocytoses have contributed to a new classification based on molecular alterations, as well as on clinical and imaging characteristics and the phenotype.
